Robust UHPLC-(ESI+)-MS/MS Method for Simultaneous Analysis of Glucoraphanin, Sulforaphane, and Sulforaphane Metabolites in Biological Samples

A UHPLC-(ESI+)-MS/MS method was developed and validated for the simultaneous determination of glucoraphanin (GR), sulforaphane (SF), sulforaphane-glutathione (SF-GSH), sulforaphane-cysteine (SF-CYS), sulforaphane-N-acetyl-cysteine (SF-NAC), in a range biological materials (digesta, cell culture medium, lysates, urine) with no sample pretreatment. The limit detection (LODs) ranged from 0.03 to 1.95 μg L–1. intra- interday variation ≤15% recoveries were ≥85%. Linearity 3.90 1000 L–1 0.06–500 3.90–1000 62.5–500 SF-CYS SF-GSH. broccoli seed powders, by determining transport metabolism SF Caco-2 monolayers, model digesta. Results indicated that 72.4% absorbed, transported, metabolized (SF-CYS, SF-GSH, SF-NAC) monolayers after 4 h incubation. This is robust time- cost-efficient suitable high throughput applications.
